ABSTRACT

AIM: To investigate the efficacy of epithelial-off accelerated corneal cross-linking(CXL)in the treatment of advanced keratoconus.METHODS: A retrospective study was performed on data collected from 32 patients(43 eyes)with advanced keratoconus who underwent epithelial-off accelerated CXL at Ningxia Eye Hospital from April 2020 to December 2021. Slit-lamp, intraocular pressure, uncorrected visual acuity(UCVA), corrected visual acuity, specular microscope, Pentacam and Corvis ST were tested before and at 1, 3 and 6mo after surgery. Preoperative and postoperative corneal condition, UCVA, best corrected visual acuity(BCVA)and the values of corneal endothelial, maximum keratometry(Kmax), thinnest corneal thickness(TCT), anterior and posterior surfaces of the cornea K1, K2, biomechanically corrected intraocular pressure(bIOP), applanation time 1(A1T), applanation length 1(A1L), applanation velocity 1(A1V), applanation time 2(A2T), applanation length 2(A2L), applanation velocity 2(A2V), highest concavity deformation amplitude(HCDA), radius at highest curvature(HCR), highest concavity peak distance(HCPD)and stiffness parameter at first applanation(SP-A1)were recorded.RESULTS: There were differences between UCVA(LogMAR; 1.06±0.49, 0.78±0.39)and BCVA(LogMAR; 0.48±0.34, 0.38±0.29)before and at 6mo after surgery(P<0.05), but there were no differences in corneal endothelial cells(2917.39±288.38 vs. 2959.19±336.27 cells/mm2, P=0.477). There were differences among Kmax, TCT, anterior surface K1 and K2 and posterior surface K1 before and after surgery(P<0.05), and all increased at 1mo after surgery then returned to preoperative level at 3mo after surgery, while there was no difference in the posterior K2. Furthermore, there were statistical significance in A1T, HCPD and SP-A1 before and after surgery(P<0.05), while there were no statistical significance in A1L, A1V,A2T, A2L, A2V, HCDA, HCR and bIOP(P>0.05).CONCLUSION: Epithelial-off accelerated CXL can prevent the progression of keratoconus within half year after surgery, and it has certain safety.

ABSTRACT

Organic anion transporting polypeptide 1B1 and 1B3 (OATP1B1/3) as important uptake transporters play a fundamental role in the transportation of exogenous drugs and endogenous substances into cells. Rat OATP1B2, encoded by the gene, is homologous to human OATP1B1/3. Although OATP1B1/3 is very important, few animal models can be used to study its properties. In this report, we successfully constructed the S knockout (KO) rat model using the CRISPR/Cas9 technology for the first time. The novel rat model showed the absence of OATP1B2 protein expression, with no off-target effects as well as compensatory regulation of other transporters. Further pharmacokinetic study of pitavastatin, a typical substrate of OATP1B2, confirmed the OATP1B2 function was absent. Since bilirubin and bile acids are the substrates of OATP1B2, the contents of total bilirubin, direct bilirubin, indirect bilirubin, and total bile acids in serum are significantly higher in KO rats than the data of wild-type rats. These results are consistent with the symptoms caused by the absence of OATP1B1/3 in Rotor syndrome. Therefore, this rat model is not only a powerful tool for the study of OATP1B2-mediated drug transportation, but also a good disease model to study hyperbilirubinemia-related diseases.

ABSTRACT

Objective@#To elucidate the clinical behavior, causes of misdiagnosis, surgical management, and outcomes of facial nerve schwannomas (FNS).@*Methods@#A retrospective review in Chinese People′s Liberation Army General Hospital from January 1, 2002 to December 31, 2015 was carried out and evaluated 110 patients with FNS, including 50 males and 60 females, aged 16-67 years old. The appropriate surgical strategy was selected based on each patient′s clinical manifestations, facial nerve function, and imaging characteristics. After surgery, patients received follow-up visits to assess their facial nerve functions, with the effect of treatment compared to the reality before surgery. The Kruskal-Wallis H test was used to distinguish between the pre- and post-operation facial nerve functions in patients who had different facial nerve functions before the operations.@*Results@#110 cases of FNS mainly presented with facial paralysis, hearing loss, tinnitus, otalgia, dizziness, and facial spasm. 20 of the cases were misdiagnosed as Bell′s Palsy, 6 were mistaken for chronic otitis media/cholesteatoma with radical mastoidectomy, 3 were mistaken for Meniere′s disease, 1 was misdiagnosed as petrous bone cholesteatoma, and 4 were mistaken for acoustic neuroma. 81.8 % (90/110) of the patients had multiple segments of the facial nerve, including the vertical segment of the facial nerve, accounting for 65.5% (72/110), followed by the labyrinthine/geniculate segment, for 61.8% (68/110), and the horizontal segment, for 55.5% (61/110). The appropriate surgical approaches were chosed based on the sizes and scopes of the tumors evaluated by imaging: transmastoid approach in 73 cases, translabyrinthe approach in 14 cases, middle cranial fossa approach in 13 cases, retrosigmoid approach in 3 cases, transmastoid-middle cranial fossa approach in 3 cases, and transmastoid-neck approach in 4 cases, with all the patients undergoing a total/subtotal resection of the tumor. Eighty-seven patients had their facial nerves reconstructed. Among them, 6 received facial nerve end-to-end anastomosis, 55 received great auricular nerve graft, and 26 were subjected to facial nerve-hypoglossal nerve anastomosis. Because of long histories, facial muscle atrophies, or other reasons, the remaining patients were not received facial nerve reconstruction. The House-Brackmann(H-B) grading scale was used to evaluate the facial nerve function pre- and post-operation. Patients with better facial nerve functions and shorter history of facial paralysis before operation would get relatively better facial nerve function. The before and after operation comparisons revealed that the recovery of the facial nerve functions in patients with H-B Ⅰ-Ⅲ was better than the improvement in patients with H-B Ⅳ-Ⅴ. The difference was statistically significant (Kruskal-Wallis H test, H=8.508, P<0.05).@*Conclusions@#The diagnosis of patients with unknown facial paralysis, hearing loss, and tinnitus should take into account the possibility of FNS. CT and other imaging examinations of the temporal bone can avoid misdiagnosis and determine the tumor size and extent of lesions, as well as provide the basis for the choice of the surgical approach. After tumors have been completely resected, facial nerve reconstruction can be performed simultaneously, according to the defect of the nerve.

ABSTRACT

Vitamin D (VD) is a multifunctional nutrient which can be either synthesized or absorbed from the diet. It plays a pivotal role in systemic calcium and phosphate homeostasis, as well as in various physiological and pathological processes. VD is converted to the active form, 1,25-dihydroxyvitamin D (1,25-D3), by cytochrome P450 2R1 (CYP2R1)/CYP27A1 and CYP27B1 sequentially, and deactivated by multiple enzymes including CYP3A4. On the other hand, 1,25-D3 is capable of activating the transcription of genes in humans, mice and rats. The vitamin D receptor (VDR)-mediated transactivation of human and resembles that known for pregnane X receptor (PXR). Activated VDR forms a heterodimer with retinoid X receptor (RXR), recruits co-activators, translocates to the cell nucleus, binds to the specific vitamin D responsive elements (VDRE), and activates the gene transcription. In mice, intestinal mRNA levels, but not those of hepatic CYP3As, were induced by administration of VDR and PXR agonists. In rats, intestinal and mRNAs were induced by 1,25-D3 or lithocholic acid (LCA), whereas hepatic , but not and , was modulated to 1,25-D3 treatment. In general, the VDR-mediated regulation of CYP3A presents species and organ specificity.

ABSTRACT

Brain injury is the main cause of neonatal death and neurological dysfunction in infants,the sequelae of which are more common,and bring serious harm to the society and the family of the patients.Therefore,early detection of brain damage in high-risk infants,evaluation of its severity and guidance for further treatment have become a hot topic in recent years.People are coming to realize the importance of continuous EEG monitoring of newborns with encephalopathy.Amplitude-integrated electroencephalogram (aEEG) is easy to operate,intuitive and easy to read.It is suitable for prolonged tracing and is suitable for continuous monitoring of brain function in neonatal intensive care unit.This article reviews the aEEG application in clinic,including the newborn with hypoxic ischemia induced brain injury,selection of appropriate patient,hypothermia treatment,neonatal convulsions,premature brain cerebral injury,premature brain maturity assessment,other high-risk infants brain damage diagnosis and efficacy evaluation.

Objective@#To elucidate the effect of taurine on neurotoxicity induced by Mn by investigating cell cycle and apoptosis in manganese exposed rats.@*Methods@#156 healthy male SD rats were randomly divided into 1 control group, 3 manganese exposure groups (10, 15, and 20 mg/kg respectively) , and 9 intervened groups based on orthogonal design, with 12 rats in each group. After 12 weeks of exposure, all rats were decapitated and striatums were removed, cell cycle was analyzed by flow cytometry, the apoptosis was detected by TUNEL, level of Mn was determined.@*Results@#The striatum apoptosis index of the 3 dose groups exposed to Mn were significantly higher than control group (P<0.05) . The striatum apoptosis index of the 9 intervened groups were significantly higher than control group (P<0.05) . 150 and 200 mg/kg of taurine could decrease apoptosis index of the group exposed to 10、15、20 mg/kg of Mn (P<0.05) . The striatum Mn content of the 3 dose groups exposed to Mn were significantly higher than that of the control group (P<0.05) . The G0/G1 proportion of the 3 dose groups exposed to Mn were significantly lower than that of the control group (P<0.05) , the S proportion of the 3 dose groups exposed to Mn were significantly higher than that of the control group (P<0.05) .@*Conclusion@#Mn could cause cell cycle arrest to S, increase level of apoptosis in striatum, to a certain extent, taurine can protect neurons from apoptosis induced by Mn.

ABSTRACT

Parthenolide is a sesquiterpene lactone derived from the plant feverfew (Tanacetum parthenium) that possesses multiple anti-inflammatory and anti-cancer properties. A specific, sensitive, accurate and precise LC-MS/MS method was developed and validated in the quantitative analysis of parthenolide in rat plasma. A liquid-liquid extraction method was used to separate the analyte and the internal standard (IS, costunolide) from plasma. A water-methanol mobile phase system was utilized in the gradient chromatographic separation. The calibration curve with good linearity (r2>0.99) was established between 2 and 128 ng·mL-1 with accuracy and precision within acceptable limits at different QC levels. High extraction recovery was achieved for both parthenolide (89.55%-95.79%) and IS (96.87%). Based on this LC-MS/MS method, the plasma stability and pharmacokinetics of parthenolide were assessed in rats. Parthenolide was proved to be very unstable in rat plasma, and was distributed and eliminated quickly in vivo, with a half-life less than 90 min. A high dose of parthenolide (80 mg·kg-1) resulted in a very low initial concentration (138.86±21.07 ng·mL-1). The systemic exposure of parthenolide (area under the curve) increased disproportionally from 40 mg·kg-1 dose group to 80 mg·kg-1 dose group. The present study may provide helpful information for the development of parthenolide as a drug candidate.

After 12 weeks of exposure, all rats were decapitated and corpus striatums were removed, activities of Na+-K+-ATPase and Ca2+-Mg2+-ATPase and content of Mn and active calmodulin were analyzed.@*Results@#The corpus striatum Mn content of the 3 dose groups exposed to Mn and 9 taurine intervened groups were significantly higher than that of the control group (P<0.05) . Active calmodulin content in 10 mg/kg manganese exposed group was significantly higher than that of the control group (P<0.05) . 150 and 200 mg/kg of taurine could decrease active calmodulin content of the group exposed to 10 mg/kg of Mn (P<0.05) . The corpus striatum activities of Na+-K+-ATPase and Ca2+-Mg2+-ATPase of the 3 dose groups exposed to Mn were significantly lower than that of the control group (P<0.05) . 150 mg/kg of taurine could increase activities of Na+-K+-ATPase of the group exposed to 10 mg/kg of Mn (P<0.05) . 150 and 200 mg/kg of taurine could respectively improve activities of Ca2+-Mg2+-ATPase of the group exposed to 15, 10 mg/kg of Mn (P<0.05) .@*Conclusion@#Mn can decrease the rats corpus striatum activities of Na+-K+-ATPase and Ca2+-Mg2+-ATPase, effect level of active calmodulin in relation to dose of Mn, to a certain extent, taurine could regulate activities of Na+-K+-ATPase and Ca2+-Mg2+-ATPase and improve the level of active calmodulin.

ABSTRACT

<p><b>OBJECTIVE</b>To explore the mechanism of neurotoxicity induced by manganese, and observe the effects on the apoptosis of neurons in rat striatum.</p><p><b>METHODS</b>SD rats were divided into four groups, six rats each group. Three dose groups were exposed to high, middle, and low level of MnCl(2). At the end of experiment, all rats of the exposed groups and control group were decapitated, their striatums were removed and the Mn content of striatum, the apoptotic morphology, ratio and ultrastructural organization were analyzed.</p><p><b>RESULTS</b>The Mn content of striatum and apoptosis index of the three dose groups exposed to high, middle, and low level of Mn were significantly higher than control group (P < 0.05). The Mn content of striatum of the three dose groups exposed to high, middle, low level of MnCl(2) and control group were 2.98 +/- 0.52, 2.75 +/- 0.37, 2.61 +/- 0.73, 0.60 +/- 0.20 respectively. The apoptosis index of striatum of the three dose groups exposed to high, middle, low level of MnCl(2) and control group were 24.83 +/- 5.98, 17.00 +/- 5.33, 15.33 +/- 2.58, 2.83 +/- 0.41 respectively, and following higher level dose, the apoptosis index increased. The nucleus of neurons in striatum become smaller, condensed, etc, and these character showed apoptosis of neurons.</p><p><b>CONCLUSION</b>Mn can result in apoptotic morphology and increase level of apoptosis in striatum. The level of apoptos varies with Mn concentration.</p>

ABSTRACT

Objective To detect the microsatellite instability (MSI) of D9S166,D9S171,D9S941, D9S942 and IFNA located at chromosome 9p21 in gastric carcinoma with high-grade intraepithelial neoplasia and normal tissue,and investigate the correlation of p14,p15,p16 gene and the gastric carcinogenesis. Methods 55 cases of gastric carcinoma with high-grade intraepithelial neoplasia and normal tissue were se- lected to detect MSI by microdissection,polymerase chain reaction,denaturing polyaerylamide gel elec- trophoresis and silver nitrate staining technology of five microsatellite markers on 9p21.Results In the infor- mative cases,total ratio of MSI in gastric carcinoma was 27 % (64/233) and it was 18% (42/233) in high- grade intraepithelial neoplasia.The ratio of MSI was significantly higher in gastric carcinoma than that in high-grade intraepithelial neoplasia (P

ABSTRACT

<p><b>OBJECTIVE</b>To explore the expression of NF-kappaB and ICAM-1 in the gas explosion wounded lung of rats and the relationship.</p><p><b>METHODS</b>Digoxin labeled NF-kappaB was used as probe. In situ hybridization was performed to detect the NF-kappaB mRNA. Immunohistochemistry was used to detect the expression of NF-kappaB and ICAM-1.</p><p><b>RESULTS</b>The levels of NF-kappaB mRNA, the expression of NF-kappaB and ICAM-1 in the wounded rats were significantly increased and reached their peak two hours after injury. Pathology of lung tissue showed that some crachea epithelium mucosae were desquamated; congestion, edema of trachea wall and infiltration of neutrophilic granulocytes were found; hemorrhage, edema and infiltration of lots of inflammatory cells were present in alveolus cells. Electron microscope showed that type I, especially type II alveolus epithelia had degeneration and desquamation.</p><p><b>CONCLUSION</b>The injury of gas explosion can activate NF-kappaB, which has close correlation with the acute injury to lung.</p>

ABSTRACT

<p><b>OBJECTIVES</b>To explore relationship between rat brain tissues hurts of gas explosion and the expression of Protein Kinase C alpha mRNA.</p><p><b>METHODS</b>Build up rat hurt model of gas explosion. In Situ Hybridization (IDH) technique was used to test Protein Kinase C alpha mRNA. Immunohistochemical Assays (IHA) was used to determine c-fos gene protein.</p><p><b>RESULTS</b>Only a little a mount expression of PKC alpha mRNA and c-fos of the control group was detected. The expression of the cerebral cortex and hippocampus of PKC alpha mRNA 24 h, 48 h and the 48 h increased obviously, and the 48 h reached the peak of expression; (t = 4.12 P < 0.01). The expression of c-fos protein of the cerebral cortex and hippocampus started to increase obviously at 0.5 h and the 4 h reached the peak, then the strength lowered gradually and the expression level came back normal level on fifth day.</p><p><b>CONCLUSION</b>The anoxia of brain tissues due to the gas explosion may promote the expression of PKCamRNA, and PKCamRNA could regulate the expression of the gene of c-fos. Both PKCamRNA and the gene of c-fos are involved in harmful processes to the nerve cells.</p>

ABSTRACT

The proposed registration algorithm based on wavelet transform is a multi-resolution block matching one, which exploits the hierarchical self-similarity of Digital Subtraction Angiography images. This method befittingly trades off between the estimation precision and computational complexity, because the registration precision is from coarse to fine, which reduces the searching complexity of motion vector. This technique has been proved that the convergence speed is exponential. The proposed method takes full advantage of the precise location both temporally and frequently which characterizes the wavelet transform. Both the matching speed and consistency are boosted without the loss of matching performance.

ABSTRACT

How to eliminate the background of other tissue and to protrude the blood vessel information is the basic requirements in DSA. Because of the complexity and non-linearity of the X-ray imaging process, the common DSA technique cannot completely remove other tissue's image which lapped over the vessels. By experiments, this paper analyses and corrects the imaging process of the actual DSA system based on the "equivalent single energy" model, and the results indicate the validity of this method.
